25068 Semen (Spermatozoa) Profile - Spec Type:
Semen - Spec Container:
Plastic container with tightly fitting lid - Pref Vol:
Entire specimen - Min Vol:
1.0 mL - Fasting:
No - Patient Prep:
The patient must not have had a fever for 3 days prior to collecting the specimen
No ejaculation for 3 days prior to each specimen collection unless instructed otherwise by the physician - Spec Collect:
Collection may be performed at home if the patient lives within 30 minutes of the testing facility
Preferred specimen is received within 30 minutes of collection, but will be accepted if received within 60 minutes of collection
Masturbation is the preferred means by which to collect the specimen as it is the method that is least likely to contaminate the sperm
Prior to collection by masturbation, thoroughly wash hands and genitals with plain soap and water. Rinse free of all soap residue and dry.
(Remember that it is very important that the laboratory receives a non-contaminated specimen, so take care to avoid touching the tip of the penis)
Do not use a condom for collection unless otherwise instructed
Do not use creams or lubricants during collection
The penis should not come in contact with the vagina during specimen collection unless otherwise instructed
Ejaculate 1 time only directly into a clean, dry, wide mouth container (obtain container from laboratory)
Entire specimen must be submitted for accurate test results (mishaps in specimen collection require collecting another specimen at a later time)
Immediately after collection, assure that lid is securely tightened and maintain container in upright position - Spec Store Transport:
Body temperature (37°C)
Tape the specimen to the body (male or female party may be responsible for transportation of specimen)
Usually the specimen container will fit between a female party’s breasts and can be supported by a bra - Spec Stability:
1 hour at body temperature (37°C) - Spec Reject:
Not received at SBMF within 1 hour of collection - Methodology:
Microscopic Examination - Use:
Infertility Studies - Clinical Significance:
Microscopic evaluation of seminal fluid for cell morphology is an important part of semen analysis. The morphological evaluation will be carefully carried out by someone familiar with seminal cytology since abnormalities may contribute to the diagnosis of male infertility or prediction of fertility success. - Reference Range:
Normal form: equal to or greater than 15%.Spermatozoa in this category exhibit an oval shaped head with a regular outline. The sperm head shows two distinct areas, the acrosome, and the postacrosomal region. The acrosome, a cap-like structure, should comprise about 40 to 70% of the anterior sperm head and stains lighter than the postacrosomal region. Borderline normal head forms and/or nearly oval heads with no gross abnormalities are regarded as abnormal. The head length is between 3 and 5 µm and the width ranges between 2 and 3 µm; the width should be between one half and two thirds of the length. If there is any doubt about the size, use an ocular micrometer. The midpiece is slender, less than one third of the width of the head, straight and regular in outline. It is aligned with the longitudinal axis of the head and is approximately 1 1/2 times the head length. No cytoplasmic droplets greater than half the area of the sperm head should be present. The tail is slender, uncoiled, free of kinks and should present a regular outline. It is approximately 45 µm in length.Immature germ cell: less than or equal to 5 per 100 spermatozoa.WBC: less than or equal to 5 per 100 spermatozoa. - Day Run:
